I want to carry out BET surface area analysis of my pretreated biomass sample to estimate how much its surface area has increased as a result of pretreatment. I ground my biomass sample in a Wiley mill with a 20MM mesh sieve and followed up with pretreatment which resulted in a reasonable delignification rate and enzymatic saccharification rate. I submitted the sample sample for BET surface area analysis and the adsorption and desorption isotherm is all over the place as pockets in between the biomass particles are interfering with the process. The technician asked me if I can bring my sample to powder to which I said it will affect my results. Anyone here faced such issues before? Please let me know what can be done to get over this problem.

The temperature that I used during vacuum degassing was 100 degree celsius for 15h. I am worried temperature as high as 300 degree celsius would affect the structure of
my biomass sample. Please share your thoughts.
Can You submit an isotherm from Your sample? It is easier to answer when we see the problem.
Najya Jabeen Poolakkalody I also think 300 C is too high temperature for degassing.The sample is most likely not a microporous one som rather short degassing can be used. If the sample is oven dry, we usually start with 140 C for 3 hours under vaccuum. Longer times could be tested.
It was said that the sampe was milled to 20 mm, how does this fit into the sample tube, the standard tube is a12 mm OD, what kind of tube is used?

The sample was ground in a 20MM mesh which is equivalent to approx. 850 micron. I did my analysis in a paid facility as my institute does not have the equipment. Since it is biomass sample, even 200 mg was occupying a lot of space in the tube. I will request them to send me the isotherm so that I can share it here. I didn't think of getting it from them since the result was unsatisfactory. Hope you would be able to help.
One rule of thumb is to have at least 20 m2 in the tube. The less mass the faster it is to degas.
