I am a student researcher, investigating whether glibenclamide can restore insulin secretion in cells with Wolfram syndrome (WFS1 knockdown). The team I am working with have been gifted some MIN6 cells with WFS1 knockdown, so in order to confirm successful knockdown we will be testing for an ER stress phenotype.
In order to produce controls for the experiment we are inducing ER stress in healthy cells. My question: before exposing these healthy cells to ER stress inducers (we will be using thapsigarin, tunicamycin and DTT), should the cells be incubated in fresh media free from serum?

I use regular medium with FBS in my ER stress experiment. You don't need to culture cell in serum-free medium for your experiment. Long time starvation will induce mildly ER stress.

Agree with Luke. Dont have to perform serum starvation for inducing ER stress.
hey! we do not remove serum to induce ER stress neither in cells or tissue in culture except for experiments which serum may interfere with your readings (LDH assays for instance). Good luck! F.
