I have to perform methylation of free fatty acids / PUFAs for GC-MS analysis. I also want to analyze oxylipins (lipids with hydroxy groups) where I perform silylation with BSFTA directly after methylation with diazomethane.
So right now, I am either using trimethyldiazomethane or diazald to produce diazomethane that is used for methylation of carboxylic groups. It is very toxic and carcinogenic.
So I was thinking of other ways for methylation in order to make the FA volantile. I already tried TMSH and lipase-catalyzed methyl esterification with non-satisfying results. Do you have any suggestions?
