measurements of leaf water potential can be easily made on monocots such as sorghum. Very quickly cover the leaf (it can be the whole leaf or part) with `saran wrap' or similar film, immediately cut the leaf with a sharp blade, place in the seal with the cut end just exposed so that it can be seen clearly with a hand lens, tighten the seal, and apply pressure slowly (tighten seal as required to stop leaks). Watch until sap just exudes from the end: record the pressure. Follow the safety instructions with the pressure chamber very carefully taking particular care to protect yourself: pressure chambers are potentially dangerous.
Since Sorghum leaves are flat, you will require a special seal (which is available with most manufacturers) for getting a system without leakage. The usual seal is for cylindrical petioles, which is not suitable for your case. In case the above type of seal is not available, a psychrometer type water potential measurement system is useful, however, the pressure chamber is very fast and easy to use.
