We do not have access to an automated gas analyzer so I am using NaOH traps in closed incubation jars. Briefly: 75 g soil (previously stored at 4 degrees) will be preincubated for 20 h before introducing NaOH traps. The basal respiration rate will be estimated by back-titration of the unreacted NaOH to determine CO2 evolved over 10 h. I am not sure about the titration step, most papers just mention that they used HCL but some methods recommend precipitating the dissolved carbonates as barium carbonate. My question is whether I can get reliable results from manual titration without precipitating the trapped CO2 with barium chloride.
