How do I densitometer a Ponceau S membrane image?

The molecular weight of my protein of interest is very close to the molecular weight of my housekeeping protein. I have tried several stripping methods, but they were all ineffective. As an alternative, they suggested using a photo of the membrane after incubation with Ponceau as housekeeping. How do I do this using Image Studio software?
