I am trying to prepare 800ml of 10% skimmed milk media, with 10% sucrose, 1% glucose, 1% casitose and 0.1% K2HPO4. I dissolved 80g of sucrose, 8gm of glucose and 8gm of casitose, and 0.8g of K2HPO4 in 400ml of Milli Q and filter sterilized it. In another bottle, I am trying to dissolve 80g of SM growth powder obtained from Himedia in 400ml of Milli Q. I autoclave it as per the manufacturers instructions (121 degrees C for 5 minutes) but each time the milk curdles. Filter sterilization does not work for me, as the filter gets clogged. Does anyone have any suggestions?
