Here is a pretty nice pharmacological study of the effect of Naspm on AMPA receptors you can look at it:

In your experiments, keep in mind that Naspm is a "use and voltage-dependent" inhibitor... It will determine the way you design your recording protocol.

we have used IEM to block CP-AMPARs. It seems quite specific and we tested that it worked well to block CP-AMPARs in retina cells that express these receptors. But we applied this drug in the bath while we recorded whole-cell from neurons.

In what kind of tissue, culture do you want to study Glua2-lacking AMPA receptors ?

Effect of NASPM 100microM in ACSF appears in 5-10 minuts at flow rate 2.5/3 ml per minute on whole cell patched CA1 pyramidal neurons EPSCs in acute or organotypic hippocampal slices. It's the same with fEPSPs.The effect is reversible after 10 minutes wash.You could have a look at this paper to have an idea of how to perform the analysis: http://www.ncbi.nlm.nih.gov.gate2.inist.fr/pubmed/26174503

You can try 100microM in the pipette for I/V experiments, wait 5 to 10 min after accessing the whole cell configuration.
