I have a "gene of interest" transformed BL21 cells, and I have harvested those BL21 cells out of that, some of them expressing protein of interest and some of them not. I doubt whether partially BL21 cells have the inclusion of body protein. Please correct me.
Hello Amaresh, I am unclear on your question. Do you mean to say that you checked for the expression of your protein from multiple colonies of BL21 and only some colonies are showing expression of that protein? Or do you truly mean that you have one culture and only some proportion of that culture is expressing your protein of interest? If it is the latter case, then that is normal as cultures will typically be genetically heterogeneous. How did you determine that only some of the culture is expressing your protein? What is your goal with this protein and why is heterogeneity an issue so long as you can obtain pure protein? More details would help.
