You can collect the uredo spores from the infected plants using the cyclone spore collector attached to a vacuum pump. Dissolve the uredo spores in distilled water and stir it. Dear Habib, To collect inoculum of rust, the stage You choose is dependent on the purpose You have. If the rust fungus is of alternate host, and if You are willing to work on alternate host, You have to collect aeciospores (transfer spores) in order to be able to successfully inoculate and infect your host plant. However, if You are due to study a rust disease on a primary host, then uredospores can be a good choice for collection as they are repeatedly and abundantly produced in a season. With the rust fungi with all stages on an only one host (Autoeceous rust fungi), the uredospores would be again the best choice due to their production in abundance in whole season.
You may just use a sucking tool as aspirator in order to collect uredospores. You may use the uredospore powder within a few days or You can store uredospores as a concentrated suspension at -20 C degree. Please consider that uredospores may release some substances in the spore suspension that may inhibit their germination in high uredospore densities. To inoculate your plants, I can offer the spraying of lower abaxial surface of plant leaves in order to be sure that You have provide enough entrances (stomates) for fungus penetration. Also, this will keep your uredospores far from direct sunlight that may otherwise desiccate it before any successful step. Then, spraying near to sunset is preferred and will favor infection because of open stomates, less evaporation rate, and coming dew formation time.
