DMBA or NMU (N-nitrosomethylurea) will induce mostly ER+ and endocrine responsive mammary tumors in rats. However, you need to pick a sensitive strain (Sprague-Dawley is probably the most widely used). There is a high incidence of V-Ha-ras mutations with NMU; these are much less common with DMBA. These drugs must be given within the appropriate developmental window or the incidence of mammary tumors is low.
The ACI rat strain is also susceptible to induction of mammary tumors by estrogen addition. There can be pituitary tumors that develop too and these can limit how long you can use the rats.
