Hi, I am using a miRCURY RNA Isolation Kit to extract exosomal RNA from 0. 5 ml serum and I got very low volume. Then I reduced the elution volume ( from 100 µL to 50 µL) in order to make it more concentrated and I increased the incubation time on the column and heated the water (37C) but still get very small RNA volume. Any advice will be appreciated.
Its bit challenging to increase the RNA concentration from 0.5ml of Serum. Which exactly RNA isiolation kit are you using ?? is it the plasma kit or the cell and plant kit ??
Secondly, it also depends on the material that you used to perform the qPCR. We use the Exiqon kit for qPCR and seems that it works really well even for small/low concentration
Hi Taral,Thank you for reply, It is Exiqone Kit(miRCURY RNA Isolation Kit) yes it is plasma and serum kit, but I have to use Taqman card for qPCR that is available in Lab.
