We're preparing NPs for the topical delivery (to avoid systemic effect). How can we determine the permeation parameters if the drug doesn't pass to the receptor chamber? What we have only the amount of drug extracted from the full thickness skin. Do we need to use only epidermis layer for this study? One more question, how can we separate the epidermis and dermis of the pig ear skin? We've tried use heat separation method (immerse the skin into 60 deg C water for 120s) but still couldn't separate it.

Since I do not know the nature if the disease and the drug you are using, my answers will be brief.
Regarding your first question, if the drug does not cross the barrier, in your case is receptor chamber, means that there is no permeation. Technically, you cannot rely on the extracted drug, the latter will be useful only if you are targeting the surface of the skin and investigating the saturation point.
My advice is better to use full skin and do not try separate its layers, using only one type of layer may lead to fake positive. You can determine the depth of drug’s location, thus you can figure out where it is stack and you can make modification on your carrier to make it cross the concerning layer.

if the drug doesn't pass to the receptor chamber, permeation does not occur...Use some permeation enhancers in your formulation e.g. TPGS, Lauric acid...
You can take section of skin using cryotome instrument..
