Your cells are already in a hyperglycemic medium-normal glucose in 5.5 mM and 4.5 mM glucose medium is ~20 mM. I believe the recommended basal media for MCF-7 cells is MEM with pyruvate and nonessential amino acids, which is 1 g/L glucose or 5.5 mM glucose.

Actually you are right, when i searched the literature i found the same molarity levels for normal and diabetic/hyperglycaemic conditions.
On the other hand it is strange that i learnt our lab (my lab mates being there much longer than me) have been using high-glucose DMEM for the MCF-7 subcultures for a very long time (by the way we didn't purchase another new cell line yet). And they even used low-glucose DMEM (1 g/L) mistakenly once, but they said it had resulted in deformations in cell shape and not a substantial growth. Right after that, when they started again the high-glucose medium treatment, the cells reached the desired manner.
