I would like to inquire whether enriching the Tryptic Soy Broth (TSB) medium with supplements such as yeast extract, meat peptone, amino acids, and vitamins can enhance the growth rate of pathogenic bacteria. Specifically, is it possible to achieve rapid or exponential bacterial growth within less than 12 hours under such conditions?
The TSB medium already has a very rich composition in these components that you suggest, since complex media are rich in amino acids, minerals and vitamins. To improve growth, it is possible to add an easily assimilable carbon source, such as glucose, but care must be taken because, depending on the microorganism, it can form organic acids that can be inhibitory to growth. Another suggestion is to use the TSB medium only to reactivate (prepare the inoculum) the microorganism and growth for the formation of biomass or to produce the molecule of interest in a defined medium with salts, vitamins and known sources of nitrogen and carbon. It really depends on what pathogenic bacteria you want to grow. TSB is often supplemented with yeast extract, sheep blood or other components to grow fastidious pathogenic and environmental bacteria.
You might investigate whether Terrific Broth would be suitable for your use. I used it years ago to enhance plasmid production in E coli.
You do need to simply try some of the supplements to see how they work. Brain Heart Infusion broth might work, I find better growth for some bacteria. I would be curious about the growth curves you are getting as I have an interest in Lag phase, but some of the published data is not reliable.
