I need a method to show that a mammalian membrane transporter such as zinc transporter has its activity after cloning and expression in yeast host cells. Is there any way to test the activity?
you could use a yeast mutant altered for Zn transport (eg. zrt1∆), which does not grow in zinc-limited medium, and check whether the expression of the mammalian transporter in these cells rescues growth on this medium.
See https://www.ncbi.nlm.nih.gov/pubmed/8637895
Beware that the heterologous expression of transporters in yeast cells can prove difficult. See https://www.ncbi.nlm.nih.gov/pubmed/16897711
Thank you very much for your helping, of course it is possible, however creating mutation is itself another project. I am seeking to find an easy approach.
