I have an acid-phenol:chloroform for RNA isolation to remove DNA, but its pH isn't acidic anymore. Can I change the pH with HCL, or will I have to prepare it again?
RNA is very critical for further applications, and it's purity is very important. Do not risk the upcoming important results because a tiny detail to prepare a new one or get over an old mix. I prefer to prepare a new one. However, if you have no other options or resources, yes, you can use Tris-HCL, but not a direct solution of HCL. In another words, to re-buffer the phenol solution to acidic state, you need to use an acidic salt, not the acid itself, you do not want to explod your Lab. There is a protocol online, if you google it, to how to raise the acidity of phenol by using the Tris-HCL. This is the safe way. Good luck
equilibrate the phenol by using the Tris-HCL as per the protocol given in maniatis. above protocol is equally good. please do not use acid or anything such because it may be harmful.
