For image processing, I am using ImageJ. I need to separate the features from the background and calculate pixel intensity of features and background. Can anybody help me in this? Thanks
You need to be aware of the phenomenon you are observing.
Is your object still or moving?
Do your objects have textures?
Do you have noise or distortion?
Do your objects have known shapes or follow a model?
Depending on the answers to the previous questions, you may pick up the best technique regardless of the software you use.

In ImageJ use:
process >> Image Calculator ...
and try to subtract the images (sample, feature mask) to get the background and do the inverse to get the feature area.

Abbas Cheddad I was trying to use mask tool, but was not successful in it. Mask an then reverse it didn't help me. I'm not sure where is the mistake You can see the attached image

You can use the Otsu threshold to decompose an input image into foreground and background parts.
