You must start from nonpolar-polar solvent with the combine of them. th general solvent usually used are hexane, ethyl acetate, chlroform, dicholoromethane and methanol.

I read that you performed flash chromatography, so you can use the same solvent system that eluted your compound(s). I'm assuming you used silica flash chromatography, so you can use silica TLC. If you used a gradient, you will need different TLC solvent systems since compounds eluting early in the gradient are non-polar compared to those eluting later in the gradient.

Generally, TLC is used to determine a solvent system for flash chromatography. Due to the nature of your mixture (not know exactly what is active), we are turning that around and using the column chromatography to determine the TLC conditions.

for testosterone extraction from testicular tissue I used the following protocols:

During extraction: I used Acetone: Ethyl Alcohol (1:1) for two times, then Ethyl Ether for three times (4, 3, and 2 ml). After evaporation, wash three times with Ethyl Ether (10, 10, and 5 ml), then add 10 ml of NaOH (1N), then separate the supernatant by separating funnel, repeat the last step two times again using 5 ml of NaOH. then wash with deionized distilled water for two times (10 and 5 ml), then separate the supernatant carefully. After evaporation it can be stored at -16 C.

Next step is the isolation of testosterone by TLC by solving with Ether (1 ml) for three times. then you can use it on TLC plate . the solvent that has been used in TLC jar was 150 ml of a mixture of Chloroform:Acetone (97:3) 

Will be better first you check in TLC plate ur active fractions with the several solvent system as: Hexan: Acetone (5:1); Hexan EtoAc (3:1); CHCl3:MeoH (4:1), EtoAc:MeoH (20:1). After findind optimal solvent system u can subject ur fraction for further isolation. For example : If ur fraction have a best separation on the TLC plate with the Hexan:Aceton (3:1), u can start  system from (10:1) for elution Column Chromatography to remove non polar compounds (such as oil, fatty acids.........) than the polarity of polar solvent should be increase step by step. Simultaneously  obtainined fraction should be run in TLC plate.
