When there are a large number of samples to read OD600 and we want to use a plate reader, how much volume should we add to each well, and does it require dilution? Do you have the complete protocol for performing it?
The issue is that the optical density take into acount the 1 cm legth of the cuvette, reading in a microplate will give you a lower OD.
You can try read sample dilutions in a cuvette and in a plate, graph the curves and get the convertion factor.
Use the most sample volume you can load in the plate, to reduce the error.

If it is a multiwell plate of 96 wells, you must add 200 uL of solution to be measured. The dilution you should test yourself, it will depend on your sample and how dense/colored/etc it is, I advise you to throw a calibration curve to evaluate the range where your absorbance is between 0.1 and 09. Otherwise you can measure your sample and a 1:10, 1:50 and 1:100 dilution to make sure it is within the reading range.
