I would like to determine if the novel drugs target peptidoglycan synthesis on gram-negative bacteria cells by staining with WGA stain, which stains the peptidoglycan layer. But the main problem here is WGA could not penetrate through the outer membrane layer of gram-negative bacteria in order to stain the peptidoglycan layer. Therefore, I would like to know if there are any other methods in permeabilized only the outer membrane of the drug-treated gram-negative bacteria cells so that the peptidoglycan layer can be stained with WGA.
