I'm isolating tissue sections and growing them in T25s and treat them. Afterwhich, i collect supernatants for analysis of released proteins. How can I check the viability of my tissues, like you would do MTT for cellular platform to check if the treatment you are adding are killing the cells or not.
Hi Ryan, how thick is your tissue? You can try fluorescent staining with propidium iodide for dead cells and calcein green for live cells. This would be easy for qualitative analysis under the microscope, for quantitative analysis you could try to quantify the flourescence or use flow cytometry after trypsination. But it depends on the type and thickness of your tissue.

you may also use the fluorecein diacetate (FDA) staining. This dye is fluorescent only in cells containing working cytosolic esterase (thus living cells). You may combine this FDA staining with propidium iodide that reflect dead cells as suggested by Ramtim.
I prefer TTC (triphenyl tetrazolium chloride) assay as all living cells (to the best of my knowledge without any exception) possess dehydrogenases. All live cells will turn red due to reducttion of triphenyltetrazolium to tiphenylformazon by dehydrogenases.
