Dear All, I want to test the antibiotic which are produced from fungi against standard bacteria. and which is the optimum temperature and media to produce the antibiotics.

You should follow NCCLS protocol. Easiest would be to grow the organism in SAB and extract culture supernatant from Stationary phase or Phase of decline. Perform Bauer Kirby disc diffusion test using all four known ATCC strains of E. coli, S. aureus, P. aeruginosa and Enterococcus faecalis. This would be the screening you need before doing purification. Good luck
I agree with Nammalwar Sriranganathan. You should grow your fungi and examine the crude extract (supernatant of culture media) as decribed in CLSI M02-A11 against the 4 known ATTC strains.
I think the oldest protocol of using culture filtrates (either crude or purified extracts through well diffusion method on bacterial lawn on agar plates and measuring the zone of inhibition.
