First, extract leaf(newly expanded) soluble proteins with 2-mercaptoethanol, glycerol & Tris.

Run the samples on SDS-PAGE.

Cut the band for Rubiso Large subunit as well as small one from the gel and elute it with formamide (incubation at 50 C).

For the calibration curve, run BSA on SDS PAGE at different concentration and follow the same procedure as for the samples.

Measure the absorbance of both samples and standards at 595 nm.
