I am also facing the similar problem with takara AAV purification kit, I have tranfected 300 ug of pAAV-CMV ,, still I am getting 10^7 vg/ul ..can anybody help me?@ Tianci Chu have you solved the problem? I will be immensely grateful if you kindly share the trouble shooting procedure.

What's the titer before you use takara AAV purification kit. What's the yield ratio of purification. In my experience, the cell state would be the key factor that influence AAV titer and activity.
