what kind of sectioning technique do you use? Which embedding medium?

Hi! We are new in that subject and area, and we tried a classic paraffin embedding and microtom sections. I must say it is not a spesific technique! As a matter of fact, I do not know any spesific technique yet.

Hi, paraffin sectioning is quite common, depending on staining. I did kryo sections for an antibody staining, recently. But this technique requires an Kryotome.

For paraffin sectioning, broken tissues mostly appear by old knifes, insufficient fixation , insufficient tissue infiltration of parafin or by warm paraffin blocks.

Do you use an automated sectioning device or by hand? If manually, have you cooled down the praffin blocks prior to slicing?

When you speak about "breaking" insufficient paraffin-infiltration comes to my mind, but as Daniel said, there are many causes.
