Searching for the correct monoclonal antibody has been quite hard, because, although in most cases companies specify the region/epitope of the antibody (e.g. extracellular domain, 1-20 aa etc), in others there is only the clone id and no more information.
In particular, I am looking for the epitope of TR75-54 or 54.7 clone of a TNFR2 monoclonal Ab. Any advice would be valuable! Thank you!
You would need to find a collaborator willing to either perform cryo-EM or traditional crystallography to determine structure of the complex, but this is time and cost heavy. An alternative is to look at what is in the literature. The receptor:TNFalpha structure is resolved, and we know from a quick search of the literature that TR75-54 competes with TNFalpha binding (oncotarget.com/article/11943/text/), so you have an approximate zip code. You could also purchase a few mAbs with specified epitopes and then perform a binding competition study to see which compete and this also provides some information as to where the clone is binding. This would be my recommendation for a starting point.
