To the best of my knowledge, it is very difficult to extract only nicotine. Since Nicotine is an alkaloid you have to extract the alkaloids using Chloroform as solvent. You can see the procedures for extraction in the links given below

You can also estimate nicotine using GC-MS method. Most importantly, the Standard nicotine should be kept away from light and solutions should be prepared immediately before use.
