Dear colleagues:
I'm trying to find out, how I can estimate significative up - regulated genes by relative quantification qPCR (RQ) using Pfaffl method. This method gives as results fold of change of transcript amount in a test condition respect a calibrator condition. Thus, if we have a fold of change in test condition of "8 times" obviously calibrator condition assume a value of 1 time. So, if we have three biological replicates and they gives in test condition 8.1, 7.8, 8.9 fold of change we get a mean of 8.27 +/- 0.57 as standard deviation. By te other hand if we look at the calibrator condition we get in every replicate a value of 1, so the mean is 1 and the standard deviation is 0.
Now my question is, how can I apply a t test if we have these values? How can I say with statistical significance that these samples are different?
