I have tried depleting neutrophils in Rag mice using Anti-ly6G (1A8; BioXCell). I have used a dose of 170-200ug/mL. It does not seem to deplete all neutrophils. It did reduce lymphocytes and neutrophils. Any suggestions will be appreciated
It seems to be a problem that anti-Ly6G antibody treatment with the 1A8 clone does not completely deplete neutrophils in C57Bl6 mice. Without knowing the details about your experiment, I assume you want to use that mouse strain. In this case there are two principal roads you could try. Firstly, optimize your antibody depletion by using, anti-GR1 RB6-8C5 instead of the 1A8 clone anti-Ly6G, which will not only affect neutrophils, but also Ly6G+ monocytes, as outlined here:
