Many thanks for the answer. but isn`t there any enumaration methods? I would like to explain this; I will inoculate the bacteria to fish. So, I will know which bacteria I have. I need to count the colinies for the next steps. Could you help me about this subject please?

Stock culture media: Marine broth/agar

After inoculation of that particular strain if you would like to determine its presence it will be better to use luminous media for initial identification. Pick the colonies that has luminous property and do 16S to confirm.

Search papers that describe the genus Photobacterium. You will directly not find the paper of Photobacterium phosphoreum as it was published in French language in another journal but since this strain is the type species of this genus you will find any paper on photobacterium suitable for your work.

You can use Conductance method for the enumeration of P.phosphoreum (Dalgaard et al., 1996. Conductance method for quantitative determination of Photobacterium phosphoreum in fish products). This method will give you quantitative determination. For the presence of P.phosphoreum you can use luminous media and 16S sequencing (directly send your culture to RefGen, Ankara, if you do not have proper infrastructure at your Faculty).
