In different protocols, different ratios of EDC and NHS are used when trying to immobilize an antibody on a surface. While some has mentioned that the EDC/NHS: Antibody ratio should be optimized, there is no reference for EDC:NHS ratio. Does anyone know?

Hi,
try with the protocols of different suppliers (such as Thermo). Usually they suggest a 2mM/5mM EDC/NHS ratio. These suggestions are for labelling of a protein or thinghs like that, so something different to what you need, but it could be a good starting point. For my experiment I use a double concentration, 4mM/10mM EDC/NHS, and for the antibody you need to try with different concentrations, looking at the best one for your application.
