There are a lot of techniques that may accomplish this, such as overlap extension PCR and adaptions of Gibson, CPEC, etc. My experience with these is limited, however, and I would appreciate any advice in how to design my approach for simplicity, cost, and efficiency. For instance, would it even be possible to delete and insert a region of this size with large primers using overlap extension? I would rather not waste time and money finding out if not!
