I was wondering how to choose the best MOI for virus production? Would it be 50% of wells infected at D5? Or 100% or else?
For a single hit kinetic you will need approximately an MOI of 5-10/cell.Example: If you want to infect 10E6 cells, you will need approximately 5-10E6 PFU /ml. If you multiply the TCID50 titer with 0.7 to get the approximate number of PFU / ml.
