The model I was able to come up is not a "robust" one. I mean, it is not done by a specific software.. It is just a simple excell file which could generate a curve (Isobologram). If you would like me to send you the file either way, just tell me.

I will add the file and a quick description of my thoughts behind it:

-To explain briefly, at our lab we work with M. tuberculosis, which has a slow rate of growth. To solve this problem, we use Resazurin (colorant agent), to reveal the assay plate post incubation. The change from blue, to pink indicates bacterial growth. In this Excel File, there is an image of a plate that I created as an example of how to apply the data. The Blue wells are water (used to keep the plate from drying, once we have to incubate for 7 days); the pink wells are the ones that showed bacterial growth; the orange ones are the ones that showed synergistic effect (when the combination of the two drugs was able to kill the bacteria). Therefore, I checked the concentration of each drug used in each marked well (orange) to plot the data... Which gave me data to apply in a Dispersion graph, considering the drug A (antibiotic in my case) being in the Y-axis, and drug B in the X-axis. Additionally, I’ve used the MIC of drug A and B to create a dotted line which would tell me that any point that is on top of the line does not have a synergic interaction. Points below the line indicate synergism and points above the line indicate antagonism. I've applied a line of tendency, that can predict, based upon the results I’ve plotted, other concentrations that could show the same effect. I don’t know if I was clear enough… If you have any doubts, you can ask me, and I’ll try to help you out. It wasn’t easy for me to “learn” how to do it, since I haven’t found any paper that explained step by step how to do it. I’ll wait for your response. PS: if you change the data according to your experiment, the graph will adapt.

To create the best isobolograms you should plot fractional MIC data of both drugs on x and y axis.
