Hi Waleed, I use Accutase to disassociate iPSCs at 37, usually in 5 min or more depending on cell lines and coating. After pippette down and up 5-10 times using serological pipettes, the cells are in single and good for cell counting

and fluorescence intensity, in 30 seconds or less

I recommend you to pick ten clonies from one culture dish, digested them with triple or trypsin 5 mins, then you can get single cells to count. now you know the cell number of ten normal clonies, and count how.many clonies in one specific dish, then you will know how many cells here in one dish.
