Instats, Graph Pad, Graph Pad Prism, Instat 3 etc. are some of the softwares which are useful in such cases. Many of them are available online freely too. You can also use the manual method using a simple graph paper also.

Dear Sara, you can calculate it simply in XL using just two points less than 50 and higher than 50 from your curve (inhibition&concentrations) you obtain an equation after fitting the data with a straight line, then just calculate your X at y = 50%.

for non linear model I think you can sigmoidal function is used to fit dose responses curves. the drug data X is in logarithmic form.

Tabulate your data in columns of % effect and Log concentration. Then select a single site competitive inhibition. The data output will give log EC50 and error, but should also give an EC50 - equivalent to IC50 for inhibition data. Best check is to make up some data and check that the EC50 is where you would expect - if not you may have selected incorrect columns or non Log data.
