I had used MTT assay for cytotoxicity and used treated cells with MTT plus DMSO as sample test, untreated cells with MTT plus DMSO as control and no cell (only medium) with MTT plus DMSO as blank.

The best way to calculate IC50 is using calcusyn software (Biosoft). you write the percentage of inhibition on the program then the program gives you all IC values.

I have no Biosoft and Graphpad software. I will use linear regression equation for obtain IC50 using R.
Percent cell inhibition can be calculated as stated in above formula but for IC50 you need to perform probit analysis. For this you need to convert concentration into log conc. and % cell inhibition into probit value through online available tables, then SPSS software will do rest of the job.
how can i calculate the percentage of inhibtion effect of two values. one value is 42 while the other is 66. how can i calculate how much percent 42 is less than 66.
