I have been trying to image my Western blot using the Fluorescent dye from Biorad called western C protein ladder (Catalog number : #161-0376). There are fluorescent bands of 2 colours : Red (emission max at 635nm) and Green (emission max at 532nm). I cannot see any signal in the image. I suspect there is something wrong with the emission filters that I am using for imaging. I have asked the company several times but failed to get complete information regarding the filters. They have just provided me with the bandpass without mentioning what filters those bandpass corresponds to. I have opened the system and checked the filters myself but there is nothing mentioned on the sides of the body of the filters.

The following is the information regarding the filters:

1. F-590 (filter is green in colour and labelled F-590) : for imaging EtBr stained DNA gel

2. 530-580nm : mentioned by the company to be used with Blue LED

3. 610-680nm : mentioned by the company to be used with Green LED

4. 670-700nm : mentioned by the company to be used with Red LED.

The colour of the last 3 filters are red (labelled as F.O58), yellow(labelled as F.O54) and black (with a red tinge observed when held against white light) (labelled as F. R66), but I donot know which one is what.

Can someone please help me out with this?
