Thank you very much Dr. Filip Konecny for this valuable paper. Its helped me a lot to understand that mechanism. specially  role of PINK1 in oxidative stress.

I have some questions if you don't mind

As you mentioned your paper that PINK1 triggers cardiomyocytes apoptosis, So Do you think that in dilated heart the mechanism would be opposite ? I mean Maybe PINK1 would trigger apoptosis lead to thin ventricular wall.

Or the mechanism could be explained as following: Absence of PINK1 increase oxidative stress which cause apoptosis to cardiomyocytes .

PINK 1-/- mice develop HT in 2 months i.e. age dependently (as per Fig 3). This phenomenon ultimately leads to pathologic cardiac HT and LV dysfunction. We observed an induction of caspase-3 proteolytic activity and significantly increased numbers of TUNEL-positive cardiomyocyte nuclei in PINK1−/− hearts compared with PINK1+/+, +/− hearts.

It can, you need to scientifically describe this using proper technique(s). Furthermore not having functional enzyme PTEN inducible kinase 1 (PINK1) in mouse cardiac ...mitochondria, please see figure S4 (supplement), is associated in mouse with an impairment of cardiac angiogenesis and fibrosis.

Not having PTEN inducible kinase 1 (PINK1), decreases protection against oxidative stress ...which leads to above mentioned..

By dilated heart do you mean (induced by e.g. volume overload, gene knock-in, out?) what caused heart (LV?) to dilate?

I meant the dilated heart that induced by inflammation which can be caused by infectious or autoimmune diseases.
