I have skeletal muscle tissue from human vastus lateralis muscle (obtained at rest, >72h exercise abstinence) before and after a 4 week HIIT program, and I am interested in changes in lipid transport proteins and lipolytic enzyme protein content. BCA assay was performed and protein concentrations were normalised. Initially, I probed for total actin, but observe a tendency to increase (possibly suggestive of increased fibre density?), I then used cyclophilin B, but one participants samples don't produce a band for it. I'm looking for ideas of alternative suitable HKPs that are unlikely to be influenced by chronic exercise or muscle hypertrophy - GAPDH, B tubulin, etc? Or would a total protein stain such as ponceau-S or coomassie be a better approach?
