Dear all, I would like to know if there is a protocol to determine the swimming speed of bacteria using microscopy. Thank you!
Microscopic video imaging facilitates precise particle movement analysis.
Procedure:
a). Cultivate motile bacteria within a suitable growth medium slowly.
b). Place a tiny droplet of bacterial suspension on microscope slide then cover it with thin coverslip situated nearby.
c). Employ a microscope featuring phase-contrast or dark-field capabilities alongside a camera operating at remarkably high speeds.
d). Capture footage really fast with high frame rates.
e). Analyze bacterial trajectories utilizing tracking software like ImageJ with TrackMate plugin or custom Python scripts.
f). Calculate speed using the equation:
v= Distance traveled / Time Elapsed
