Currently I am using HIF1a ab from CST (D5F3M) to probe for HIF1a levels. However, in my hypoxic samples I did not see any induction of HIF1a. I then attempted to rescue the HIF1a from proteasomal degradation by applying 10 uM of Mg132 over a period of 3 hours, however it did not show up in my blot as well. My questions are:

1) is 10 uM of Mg132 for 3 hours too little?

2) It seems like people prefer BD for humans and novus for mouse HIF. Should I switch to their antibodies, and if so, which ones have given you the best result?
