some times in laboratory, LB media become cold, in this situation i dont know whether could I add antibiotic on it or no? because in normal condition i must add antibiotic when it is nearly hot.

Well you are confusing two different things, you put the ampicillin in the LB Agar, when it is nearly hot so as to ensure that the heat doesn't degrade the antibiotic and not that the heat activates the antibiotic. Furthermore, you add it at such a temperature that the antibiotic does not get degraded while the agar doesn't solidify. When it comes to the broth, you can add the antibiotic when it comes to the room temperature if the broth had been kept at a colder temperature. Adding the antibiotic once the broth comes to the room temperature should pose no problem, but do make sure that the broth hasn't been contaminated. Hope this helps.
Thanks, I know the difference between agar and media, but if I prepare LB broth and put it in refrigerator(4 degree centigrade) for 24 hours, could I bring it out in room temperature and add ampicilin now?
Thanks, I am sure now that if I fridge LB broth without ampicilin, I can add ampicilin on it some days later in room temperature to have ampicilin.thats ok?
