1. For particular system, one has to develop dissolution method by considering application, volume, rpm, medium, paddle/basket/diffusion cell etc. Ultimately we have to perform dissolution development method.
2. Replacement of the volume we withdrawn with the fresh buffer must be calculated in drug release.
Actually, there are many ways to perform that, one of them through HPLC or mass spectrophotometry.

As many times you diluted the medium to get your results in a linearity (Lambert Beer's Law) range. So depending on dilution you made, you have to calculate dilution factor.

َDetermining the volume of the release media depends on many factors, including:
the method used to study drug release (sample and separate, dialysis membrane, etc.), drug solubility in release medium, stirring speed, composition of release medium (water, PBS, etc.) and so on. The method used for analysis (HPLC, UV-Vis, etc.) is also important. for example, if the medium/released-drug ratio is too high, it may be difficult to detect by UV-Vis spectrophotometer. Therefore, according to the influencing factors, the volume of the medium should be selected. In addition, release data are usually reported as a percentage of cumulative release over time, so changing drug concentrations by replacing the fresh medium does not adversely affect the final result.
