So by this technique , establish first the aminoacid separation pattern & then use little more concentrated solvent system than the TLC for the column chromatography to separate this aminoacid from their mixtures. Separate them from what? Other aminoacids? Strictly speaking, lysine and Cbz-lysine have neither methyl, nor hydroxyl groups. Do you mean lysine-rich peptides? Methyl groups are very hard to react.
