We've been using the AdvanceBio SEC column 120A 1.9um 2.1x150mm PEEK with an Agilent LC QTOF but recently we have been problems with this column and we were not able to perform any runs.
Hello. The best SEC columns that I have used are the TSKGel PW and SW lines from Tosoh and the Ultrahydrogel line from Waters. This is based on longevity, low shedding, applicability to wide chemistries, good pH stability range in the case for PW and Ultrahydrogel. The SW line from Tosoh is better suited for globular polymers such as antibodies and many ADCs, and also can handle higher organic phase.
Some suggestions below. Check the brochures online to make sure the pH range, %organic solvent tolerance, and temperature match your needs. Often cause for SEC column deterioration is running at inadequate pH and %organic phase. And as always with SEC, ramp flow rates up OR down slowly over time, and let them run overnight at low flow rate before first use to "polish" them.
TSKgel G3000SW
TSKgel SuperSW3000
TSKgel UP-SW3000-LS (specialized for mass spec and light scattering detectors)
I strongly recommend Waters XBridge Premier Protein SEC Column with MaxPeak Premier Protein SEC Guard as the far best SEC column (couple) in terms of providing resolution, reproducibility, and stability...The main issue with this column is that you should strictly follow the user guide and need to keep your LC system and all tubings sterilized. Any microbial growth leading to detrimental effects on the SEC column (experienced, unfortunately)...I also suggest you use the guard to extend your column lifetime which is a bit expensive to purchase.
Thank you so much! I am not sure we will be able to sterilise the tubings due to the type of analysis we are doing on our MS instrument but I will keep all your inputs in mind and I will definitely check those columns out! Petroslav Chipinski İsmail Emir Akyildiz
