We've been trying chemical transformations of plasmids into E. coli (DH5alpha) using a standard chemical transformation/heat shock protocol and not having a lot of success. Even the pUC19 control cells did not grow. Is there any specific steps that are vital to be accurate in? We timed the heat shock in the waterbath to the second, and are very confused why none of the transformations work.
