What cell sample will you be using?

If a mixed sample (e.g. whole blood), which cell type(s) are you interested in?

I will work with the whole blood of patients with hereditary spherocytosis. I prepared an erythrocyte suspension. But I could not run on the device.

Thanks for the additional information. I am unclear what you mean that you could not run on the instrument, but assume that you could not identify the erythrocyte population.  

It is best to use log scale for the forward scatter and side scatter as this shows the erythrocytes as a visually discrete population well separated from platelets and leukocytes.  Hope this helps.
