Methanolic and ethanolic extraction is a best solvent extraction method to isolate higher amount of phytochemicals. After extraction, using some chromatography techniques (TLC and Etc) to isolate the Tannin and Alkaloids.

You have to start with non-polar solvent e.g. n-hexane or petroleum ( defatting) then use a solvent with higher polarity,then ethyl acetate, methanol. methanol-water 70:30. Check all fractions with TLC.Finally separate the component of each fractions using chromatographic techniques like preparative TLC , column ,the best one is MPLC direct and everce.

It is better to extact using ultrasound bath( sonation) at room temp. instead of traditional methods.
